摘 要:目的观察异氟烷麻醉对发育期大鼠神经元凋亡及远期学习记忆能力的影响,明确两者的关系。方法 7日龄SD大鼠32只,随机分为对照组(C组)和异氟烷麻醉组(A组)。A组大鼠在35℃恒温下1.5%异氟烷麻醉6 h,C组自主吸入空气6 h。每组取3只大鼠,行动脉血气分析。每组各另取3只大鼠用免疫组织化学法检测大脑皮层activeCaspase-3阳性神经元,计算阳性率。其余大鼠继续饲养至出生后60 d,行Morris水迷宫实验。后用免疫组织化学法检测大脑NeuN表达,比较两组大鼠皮层神经元密度。结果两组大鼠动脉血气分析结果差异无统计学意义。A组大鼠皮层active Caspase-3阳性神经元数量明显多于C组(P<0.05)。水迷宫实验结果显示,两组大鼠在寻台潜伏期、目标象限停留时间和穿越平台次数等指标上的差异均无统计学意义。两组大鼠皮层神经元密度相当。结论 1.5%异氟烷麻醉6 h能诱导发育期大鼠皮层神经元凋亡增加,但是对远期学习记忆能力没有影响。Objective To investigate the effects of neonatal isoflurane exposure on neuron apoptosis,long-term learning and memory in developmental rats.Methods Seven-day-old SD rats were randomly assigned to receive 6 h of 1.5% isoflurane exposure in 35 ℃(anesthesia group),or room air(control group).Arterial blood gas was measured and active Caspase-3 positive neurons in the brain cortex of rats were detected by using immunohistochemistry after 6 h of exposure in each group.Spatial learning and memory were evaluated through Morris water maze 53 days after exposure.The neuron density was assessed in brain sections stained with NeuN.Results No difference was found in arterial blood gas analysis between two groups.Active Caspase-3 positive neurons were increased significantly in brain cortex after 6 h of 1.5% isoflurane exposure.But spatial learning and memory were similar in two groups in adulthood,regardless of neonatal isoflurane exposure.The neuron density was also similar in brain cortex in two groups.Conclusion Isoflurane exposure for 6 h led to increased cortical neuron apoptosis,but no significant reduction of spatial learning and memory and neuron density was observed.
