摘  要:目的:比较不同产地延胡索药材的鲜品、水煮品、醋煮品和酒炙品的高效液相指纹图谱,探索延胡索鲜品及其炮制品间的区别。方法:将各产地的延胡索鲜品进行水煮、醋煮和酒炙后,进行甲醇加热回流提取,采用RP-HPLC定量分析法,Agilent TC-C18(250 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-0.1%磷酸溶液(三乙胺调pH 6.0)梯度洗脱,流速1 ml/min,检测波长280 nm,柱温30℃。数据处理采用中药色谱指纹图谱相似度系统软件,国家药典委员会2004A版。结果:各产地的鲜品及炮制品间的相似度均在0.9~1之间。鲜品炮制后多数成分的含量均有所升高,东阳药材炮制后增加了一个色谱峰。计算5种已知有效成分在18批鲜品药材及炮制品中的含量范围。结论:鲜品炮制前后峰的数量变化不大,只是成分的含量有增减;采用指纹图谱测定已知成分的含量范围的方法可以在延胡索药材内在质量的研究与控制中广泛应用。Objective: to compare the HPLC fingerprint of Rhizoma Corydalis with processed Corydalis from different places of produce and collecting seasons to determine the difference between Rhizoma Corydalis and processed Corydalis.Methods: The corydalis from different places was processed by water-boiling,vinegar-boiling and vine-frying.Then the samples were extracted by methanol.Agilent TC-C18(250 mm×4.6 mm,5 μm) was used as stationary phase with a temperature of 30 ℃ and a solution of acetonitrile and 0.1% phosphonic acid(pH 5.3 with triethylamine) as the mobile phase with linear gradient elution.The flow rate was 1 ml/min.The detective wavelength was 280 nm.The HPLC fingerprints were analyzed by similarity software of fingerprint of traditional Chinese medicine(2004A).Results: The similarity of Rhizoma Corydalis from different places and collecting seasons and their process were all between 0.9~1.The contonts of most active ingredients increased after the corydalis processed.A new ingredient appears after corydalis from Dongyang processed.The content range of the five active ingredients were calculated in Rhizoma Corydalis and processed Corydalis from 18 places.Conclusion:The contents of ingredients increased or decreased after Corydalis procesed and the amount of ingredients changed a little.The method of determining the content of known ingredients by fingerprint can generally applied in the study and control of the quality of Rhizoma Corydalis and its patent medicine.
