摘　　要：以企鹅珍珠贝(Pteria penguin)闭壳肌为样品,利用高盐+十二烷基硫酸钠(SDS)除去蛋白和多糖,用低盐+十六烷基三甲基溴化铵(CTAB)进一步纯化DNA,建立一种贝类基因组DNA简便、安全、经济的提取方法。改进后的SDS-CTAB法提取的DNA纯度较高,无蛋白质和RNA污染;酶切和AFLP实验验证表明,所提取DNA可满足酶切分析和分子标记等实验的要求。By using the adductor muscle of Pteria penguin as an experimental material,a simple,economical and safe Molluscs genetic DNA extraction method was established.This method uses high salt to remove proteins and polysaccharides in a sodium salt（SDS） extraction buffer.Further DNA purification is achieved by using a low salt with cetyltrimethylammonium bromide（CTAB） treatment.The DNA extraction by the improved SDS-CTAB method has high-purity,non-protein and RNA pollution.The DNA extraction method is validated by restriction enzyme digestion and AFLP analysis,and the results showed that DNA is good enough for the further biotechnology experiment such as digestion by restriction enzyme,molecular marker and so on.
