摘 要:采用PCR-SSCP方法对寿光鸡CAPN1基因进行多态性检测,并分析它们的多态性与部分肉质性状的关系。结果表明,含有CAPN1基因外显子5的引物CE5扩增的座位在群体中检测到3种基因型AA、AB和BB,基因型频率分别为0.266、0.520和0.214;等位基因A和B的频率分别为0.524和0.476。CE8引物扩增的座位在群体中也检测到3种基因型AA、AB和BB,基因型频率分别为0.378、0.510和0.112;等位基因A和B的频率分别为0.633和0.367。关联分析结果表明:含有CAPN1基因外显子5的引物CE5座位对肌内脂肪含量和腿肌蒸煮损失存在显著性影响(P<0.05),CE8座位对胸肌蒸煮损失存在显著性影响(P<0.05)。PCR- SSCP method was used to detect the polymorphism of CAPN1 gene in Shouguang chicken,and its relationship with some meat quality traits of Shouguang chicken was also analyzed. Three genotypes AA, AB and BB were detected at CE5 locus. Frequencies of three genotypes were 0. 266, 0. 520 and 0. 214, respectively. Frequencies of allele A and B were 0. 524 and 0. 476, respectively. Three genotypes AA, AB and BB were detected at CE8 locus. Frequencies of three genotypes were 0. 378, 0. 510 and 0. 112,respectively. Frequencies of allele A and B were 0. 633 and 0. 367, respectively. Statistical analysis indicated that the CE5 locus showed a significant effect on intramuscular fat content and cooking loss of leg muscle (P〈0.05). The CE8 locus showed a significant effect on cooking loss of chest muscle (P〈0.05).
