HPLC法测定活血安痛酒中芍药苷、丹酚酸B的含量

摘　　要：目的:采用HPLC测定活血安痛酒中芍药苷、丹酚酸B的含量。方法:色谱柱为Kromasil C18柱(250 mm×4.6 mm,5μm)。流动相:测定芍药苷为甲醇-1%冰醋酸水溶液(30∶70),检测波长为230 nm;测定丹酚酸B为甲醇-乙腈-1%冰醋酸水溶液(27∶13∶60),检测波长为286 nm。柱温:25℃;流速:1 ml/min。结果:芍药苷、丹酚酸B的进样量分别在0.24～2.40μg(r=0.999 7,n=6)、0.32～3.20μg(r=0.999 4,n=6)的范围内与峰面积呈良好的线性关系。芍药苷的平均回收率为99.26%,RSD为1.58%;丹酚酸B的平均回收率为98.22%,RSD为2.16%。结论:该方法简捷、稳定、可靠,拟作为有效控制活血安痛酒质量的参考指标。Objective： To establish a method for determination of Paeoniflorin and Salvianolic acid B in Huoxue Antong Wine by HPLC.Method： The sample was separated on Kromasil C18（250 mm×4.6 mm,5 μm） column,the mobile phase was methanol-1% glacial of water（30∶70） for Paeoniflorin at a flow rate of 1.0 ml/min and the detection wavelength was 230 nm and the column temperature was 25℃.Another was methanol-acetonitrile-1% glacial of water（27∶13∶60） for Salvianolic acid B at a flow rate of 1.0 ml/min and the detection wavelength was 286 nm and the column temperature was 25℃.Results： The calibration carves were linear in the range of 0.24-2.40 μg for Paeoniflorin（r=0.999 7,n=6） and 0.32-3.20 μg for Salvianolic acid B（r=0.999 4,n=6）.The average recovery of Paeoniflorin was 99.26% with RSD of 1.58%;the average recovery of Salvianolic acid B was 98.22% with RSD of 2.16%.Conclusion： The method is simple,accurate,reproducible and can offer references for the quality control of Huoxue Antong Wine.
