路氏滋燥汤对干燥综合征小鼠颌下腺细胞因子IFN-γ、IL-10的影响

摘　　要：目的观察路氏滋燥汤对干燥综合征小鼠颌下腺细胞因子干扰素-γ(IFN-γ)、白细胞介素-10(IL-10)的影响,探讨路氏滋燥汤治疗干燥综合征的作用机制。方法采用百日咳疫苗加完全弗氏佐剂及同种鼠颌下腺抗原联合诱导干燥综合征动物模型,以路氏滋燥汤灌胃治疗,并以生理盐水灌胃作为对照,以免疫组化法测定各组小鼠颌下腺中IFN-γ、IL-10水平。结果与模型组相比,路氏滋燥汤高、低剂量组小鼠治疗后颌下腺IFN-γ表达率显著降低(P<0.01),IL-10表达率显著提高(P<0.01)。与路氏滋燥汤低剂量组相比,高剂量组IFN-γ表达率显著降低(P<0.01),IL-10表达率差异无统计学意义。结论路氏滋燥汤能抑制实验性干燥综合征小鼠IFN-γ的分泌,同时促进IL-10的分泌,是其治疗干燥综合征的作用机理之一。Objective To observe the effect of Lu′s zizao decoction on cytokine IFN-γ and IL-10 in the salivary gland of mice with Sjogren′s syndrome（SS）,and investigate the mechanism of Lu′s zizao decoction in the treatment of Sjogren′s syndrome.Methods SS animal model was induced by antipertusis vaccine,complete Freund′s adjuvant and salivary gland antigen of mice in the same race.Lu′s zizao decoction was adopted in the treatment with gastric infusion.The gastric infusion with normal saline was taken as the control.Immunohistochemical method was used to measure the levels of IFN-γ and IL-10 in the salivary gland of each group.Results Compared with control group,the level of IFN-γ decreased and the level of IL-10 increased significantly in the salivary gland of the mice in Lu′s zizao decoction high and low dose groups.Conclusion Lu′s zizao decoction can inhibit the generation of IFN-γ and promote the generation of IL-10 in experimental mice with Sjogren′s syndrome,which can be taken as one of the mechanisms for the treatment of Sjogren′s syndrome.
