摘 要:目的:探索大黄虫丸对肺纤维化形成阶段的干预作用机制。方法:将48只普通级健康wistar大鼠用平阳霉素复制肺纤维化模型大鼠后,分为正常对照组、模型对照组、大黄虫丸组和强的松对照组。大黄虫丸剂量1.2 g.kg-1,从第14 d开始按15 mL.kg-1体重ig,造模28 d后处死,测定、比较各组大鼠血清和支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中超氧化物歧化酶(superoxide dismutase,SOD)活性及丙二醛(malondialdehyde,MDA)含量。结果:模型组大鼠血清及BALF中SOD活性低于正常对照组、MDA含量高于正常对照组,大黄虫丸组SOD活性均高于模型组,MDA含量均低于模型组;大鼠BALF中SOD活性、MDA含量与血清SOD活性、MDA含量呈正相关。结论:大黄虫丸有抑制模型大鼠肺及血清中SOD活性下降及MDA含量增高的作用;血清SOD活性和MDA含量可间接反映肺部脂质过氧化与抗氧化水平。Objective:To explore the mechanism of intervention effect of Dahuang Zhechong Wan on the formation stage of pulmonary fibrosis in rats.Method:The pulmonary fibrosis model was induced by bleomycin A 5(BLMA 5).Rats in herbal groups were administrated with Dahuang Zhechong Wan(consisted of rhubarb and eu-polyphaga) from the day 14,and sacrificed on day 28.Then superoxide dismutase(SOD) activity and the contents of malondialdehyde(MDA)in serum and in bronchoalveolar lavage fluid(BALF) were determined and analyzed.Re-sult:The SOD activity in serum and BALF in model group were lower than those in control group.the MDA contents in serum and BALF in the model group were higher than those in the control group.SOD activity in Dahuang Zhe-chong Wan group were higher than those in the model group,MDA contents in Dahuang Zhechong Wan group were lower than those in the model group.Conclusion:Dahuang Zhechong Wan had the functions of restraining the de-cline of the SOD activity and the increase of MDA contents of the mode with pulmonary fibrosis and serum.The SOD activity and MDA contents of serum indirectly reflected the level of lipid peroxidation and antioxidation of the lung.
