摘 要:为了克隆新城疫病毒LaSota株HN基因,构建原核重组表达质粒pBI121-HN,使HN基因在苜蓿中表达,将新城疫病毒LaSota株接种于鸡胚,培养、纯化后收获鸡新城疫病毒,提取病毒RNA,经反转录后通过PCR扩增HN基因,克隆得到重组质粒pBI121-HN,并其通过电转化导入根癌农杆菌中,以根癌农杆菌为介导将HN基因导入苜蓿细胞。最终成功构建了重组表达质粒pBI121-HN,并通过根癌农杆菌转染苜蓿细胞将其在苜蓿中成功表达。本试验为家禽的植物性苜蓿疫苗的研究奠定了基础。Cloning newcastle disease virus HN gene of LaSota strain,constructing recombinant plasmid pBI121-HN,and expressing HN gene in alfalfa,Newcastle disease virus LaSota strain was inoculated in to chicken embryo,and newcastle disease virus was cultured purefied and harvested.The viral total RNA was extracted from the Lasota virus,and the HN gene was synthesized using the AMV reverse transcriptase.Recombinant plasmid pBI121-HN was cloned,then electrotransformed into Agrobacterium tumefaciens which contain pBI121-HN and introduced into alfalfa.The result showed that recombinant plasmid pBI121-HN was cloned and expressed in alfalfa to which pBI121-HN was transfected through Agrobacterium tumefaciens.The experiment established foundation for poultry alfalfa vaccine of research.
