紫云英根瘤菌培养基的选择与优化

摘　　要：以中慢生华癸根瘤菌13004#菌株为供试菌株,进行YMAS、M、63#、TY和BSE 5种培养基筛选、碳氮源利用试验和正交设计试验。结果表明:该根瘤菌株在YMA中生长良好,最佳碳源为蔗糖,最佳氮源为酵母粉,最佳培养基成分配方为:蔗糖10 g/L,酵母粉3 g/L,K2HPO4 0.5 g/L,MgSO4.7H2O 0.02 g,NaCl 0.1 g/L,CaSO4 0.2 g/L,Rh微量元素液4 mL/L,pH 6.8～7.0。The middle-low growing strain Mesorhizobium huakuii 13004# was taken as experimental strain to conduct medium selection with five media（YMA medium,SM medium,63# medium,TY medium and BSE medium）,utilizing experiments of carbon source and nitrogen source and orthogonal experiment.The results showed that the Mesorhizobium huakuii 13004# grew greatly in the YMA medium;the optimum carbon source is sucrose;the optimum nitrogen source is yeast powder;the optimum ingredients of medium is as follows： sucrose 10 g/L,yeast powder 3 g/L,K2HPO4 0.5 g/L,MgSO4·7H2O 0.02 g/L,NaCl 0.1 g/L,CaSO4 0.2 g/L,microelement liquid of Rh 4 mL/L and pH 6.8～7.0.
