摘　　要：目的研究3-己基异香豆素的合成方法。方法以邻碘苯甲酸为原料,经酯化、Sonogashira交叉偶联以及亲电环化反应三步合成3-己基异香豆素。结果重点考察了催化剂、配体和碱等因素对Sonogashira交叉偶联反应的影响,以及催化剂、溶剂和温度等因素对亲电环化反应的影响,在最佳的反应条件下,反应总产率为79.5%,中间产物和目标产物的结构经NMR和MS得以确证。结论该法操作简单,收率较高,产物立体选择性好。Purpose To study on the synthesis of 3-hexyl-1H-isochromen-1-one.Methods 3-hexyl-1H-isochromen-1-one was synthesized via esterification reaction,Sonogashira coupling and annulation reaction,using 2-Iodobenzoic acid as the initial materials.Results Emphasis was placed on studying the influence of catalyst,ligand and base on Sonogashira cross-coupling reaction and the influence of catalyst,solvent and reaction temperature on annulation reaction.Under optimum conditions,the yield of the product was up to 79.5%,and the structures of intermediate and target product were identified by NMR and MS spectra.Conclusion The reaction was readily performed with good yield and good selectivity under convenient operations.
