摘 要:目的观察羧甲基壳聚糖钙对CCl4损伤小鼠丙氨酸转氨酶(ALT)和天门冬氨酸转氨酶(AST)活性及肝脏抗氧化能力的影响。方法将小鼠分为正常对照组、CCl4肝损伤模型组、羧甲基壳聚糖钙低、中、高剂量实验组,灌胃给予受试物。连续20 d后腹腔注射CCl4,24 h后采血测ALT、AST活性,同时取肝脏制匀浆,用试剂盒测定丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)值。结果羧甲基壳聚糖钙能显著降低ALT和AST活性,抑制脂类过氧化,提升SOD活性和T-AOC水平,对GSH-Px无显著影响。结论羧甲基壳聚糖钙对CCl4引起的急性肝损伤小鼠具有一定的抗氧化能力和肝保护作用。Purpose To observe the effects of carboxymethyl chitosan calcium(CCC) on activities of serum ALT and AST,and antioxidant activity of liver in mice injured by CCl4.Methods Mice were randomly divided into 5 groups,including normal group,CCl4 group and experimental groups treated with CCC in three different doses.CCC was administrated to mice i.g.once a day.Twenty days later,24 hours after the administration of CCl4,eyeball blood was collected and serum was separated for determination of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activities.Then the animals were sacrificed and livers were dissected out.Maleicdialdehyde(MDA),total antioxidative capacity(T-AOC),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px) activities in liver were measured by using assay kit.Results CCC significantly reduced the activities of serum ALT and AST,markedly decreased the level of MDA and increased the activities of SOD and T-AOC.However CCC had no significant effect on GSH-Px.Conclusion The results indicated that pretreatment with CCC could efficiently improve the antioxidant activity and protect mice from CCl4 induced liver damage.
