摘 要:目的:运用反相高效液相色谱法同时测定不同产地龙胆中龙胆苦苷(GPS)和2’-(2,3-二羟基苯甲酰)-獐牙菜苷(DHBS)的含量。方法:色谱柱为Ultimate XB-C18(150 mm×4.6 mm,5μm);甲醇-乙腈-水为流动相,梯度洗脱,检测波长为270 nm和215 nm,柱温为25℃,流速为1.0 ml/min。结果:龙胆苦苷和2’-(2,3-二羟基苯甲酰)-獐牙菜苷的线性范围分别为0.0201 5~5.038μg(r=1)和0.00512~0.409 6μg(r=0.999 8),平均加样回收率分别为100.4%(RSD 2.72%)和102.2%(RSD 1.99%)。结论:本法准确、简便、重现性好。龙胆的种间(四基源药材)的龙胆苦苷和2’-(2,3-二羟基苯甲酰)-獐牙菜苷的含量有较大差异。Objective: To determine the contents of gentiopicroside(GPS) and 2'-(2,3-dihydroxybenzoyl)-sweroside(DHBS) in Radix Gentianae from different sources by RP-HPLC.Methods: Ultimate XB-C18(150 mm×4.6 mm,5 μm) column was adopted at 25℃,methanol(A)-acetonitrile(B)-water(C) gradient system as mobile phase with flow rate of 1.0 ml·min-1 was used,and the detecting wavelength was 270 nm and 215 nm.Results: The linear ranges of assay of GPS with correlation coefficient being 1 and DHBS with correlation coefficient being 0.999 8 were 0.020 15~5.038 μg and 0.005 12~0.409 6 μg,and the average recovery rates were 100.4%(RSD=2.72%) and 102.2%(RSD=1.99%) respectively.Conclusion: This method is simple and reproducible and the analytical results are reliable and accurate.The contents of GPS and DHBS are significantly different in Radix Gentianae from different species and sources.
