摘  要:采用高效液相色谱法测定金银花不同部位(花,叶,茎的表皮和茎)中木犀草素及其苷的含量.应用AglientZorbax SB-C18色谱柱,以甲醇(φ甲醇=0.02%)-磷酸溶液(φ磷酸=50%)为流动相测定木犀草素,以乙腈(ρ乙腈=0.5%)(A)-冰醋酸溶液(B)按VA∶VB=1∶9,梯度洗脱至3∶7为流动相测定木犀草苷,流速0.3 mL/min,柱温30℃,检测波长350 nm.分析结果表明:木犀草素存在于金银花的花,叶和茎的表皮中;木犀草苷存在于金银花的花和叶中.该方法操作简便、结果准确、专属性强,可用于金银花植物中木犀草素及苷的含量测定.HPLC method was developed to determine the contents of luteolin and luteoloside in different parts of Lonicera japonica(flower,leaf,epidermal stem and stem).Samples were determined by HPLC on Aglient Zorbax SB-C18column.Luteolin was isolated using methanol(φ=0.02%)-H3PO4(φ=50%) as the mobile phase,the mobile phase of isolating luteoloside was 0.5% acetonitrile(A) and-acetic acid(B),gradient elution V(A)∶V(B) from 1∶9 to 3∶7.The flow rate was 0.3 mL/min,column temperature was 30 ℃,UV detection wavelength was at 350 nm.The results are that luteolin exists in flower,leaf and epidermal stem and that luteoloside only exists in flower and leaf.These methods were simple,accurate,uied.It can be suitable for the determination of the contents of luteolin and luteoloside in Lonicera japonica plant.
