理脾调脂胶囊对血脂失调症大鼠ApoE、ApoCⅢ基因表达的影响

摘　　要：目的研究理脾调脂胶囊对血脂失调症大鼠ApoE、ApoCⅢ基因表达的影响,探讨其调节血脂代谢的作用机制。方法 Wistar大鼠随机分为正常组、模型组、理脾调脂胶囊治疗组(简称治疗组)、血脂康对照组(简称对照组),造模4周后,治疗组大鼠每日灌胃理脾调脂胶囊,对照组每日灌胃血脂康,用药8周。实验结束时,酶法测定大鼠血清TC、TG浓度,沉淀法测定大鼠血清HDL-c、LDL-c;实时定量PCR法(Real-time PCR)检测大鼠肝脏ApoE、血浆中ApoCⅢmRNA的表达。结果与正常组比较,模型组大鼠血清中的TC、TG、LDL-c均显著升高(P<0.01),HDL-c降低(P<0.01),ApoEmRNA的表达量降低(P<0.05),血浆ApoCⅢ基因表达显著升高(P<0.01);与模型组比较,治疗组和对照组大鼠血清中TC、TG、LDL-c均降低(P<0.01),治疗组大鼠血清HDL-c升高较为明显(P<0.01),ApoEmRNA的表达量升高较为显著(P<0.01),血浆ApoCⅢ基因表达显著降低(P<0.01);与对照组比较,治疗组大鼠血清中TC、TG、LDL-c均降低(P<0.05),HDL-c升高(P<0.01),血浆ApoCⅢ基因表达显著降低(P<0.01)。结论理脾调脂胶囊能显著提高实验性血脂失调症大鼠肝脏ApoEmRNA的表达,并降低血中ApoCⅢmRNA的表达,具有调控血脂代谢的作用。Objective To observe the effect of ＂Lipi Tiaozhi Capsule＂ on expressions of ApoE and ApoCⅢmRNA in dyslipidemia rats.Methods Wistar rats were randomly divided into normal group,model group,treatment group and Xuezhikang control group.After modeling for 4 weeks（except normal group）,the treatment group was given ＂Lipi Tiaozhi Capsule＂ orally once a day.After 8-week treatment,the concentration of TC and TG in serum was detected by enzymatic method,the concentration of HDL-c and LDL-c by precipitation,and the expressions of ApoEmRNA in the hepatic tissue and ApoCⅢmRNA in serum of the rats from each group by Real-time PCR.Results Compared with the normal group,the serum content of TC,TG,LDL-c in model group was increased significantly（P0.01）;the serum content of HDL-c was decreased（P0.01）;the expression of ApoE mRNA was decreased（P0.05）,and the expression of ApoCⅢ in blood plasma was increased significantly（P0.01）.Compared with the model group,the serum content of TC,TG and LDL-c in treatment group and control group were decreased（P0.01）;the serum content of HDL-c was increased significantly（P0.01）;the expression of ApoE mRNA was increased significantly（P0.01）;the expression of ApoCⅢ in blood plasma was decreased significantly（P0.01）.Compared with control group,the TC,TG and LDL-c in treatment group were decreased（P0.05）,HDL-c increased（P0.01）,and the expression of ApoCⅢ was decreased significantly（P0.01）.Conclusion ＂Lipi Tiaozhi Capsule＂ can increase the expression of ApoEmRNA and decrease the expression of ApoCⅢ mRNA to regulate blood lipid.
