摘  要:目的:测定8-氯腺苷(8-Cl-A)及其代谢物8-Cl-Ad和8-Cl-Ⅰ在犬血中的药动学。方法:用RP-HPLC测定犬血中和组织中8-Cl-A及其代谢物。结果:犬单剂量和多剂量静脉点滴8-Cl-A后,计算得到8-Cl-A的代谢物8-Cl-Ad的药动学参数。结论:8-Cl-A在犬体内迅速代谢和消除血中未出现蓄积性现象,8-Cl-A在犬体内主要以8-Cl-Ad代谢物分布于主要脏器组织中。OBJECTIVE:To determine the pharmacokinetics of 8 Cl A and its main metabolited,8 Cl Ad and 8 Cl I,in dogs.METHOD:A simple and accurate RP HPLC method was used to determine 8 Cl A and its metabolites in blood and tissues.RESULTS:The main pharmacokinetic parameters of 8 Cl Ad in dogs were obtained after single dose and multiple dose iv infusion of 8 Cl A in dogs.CONCLUSION:The parent compound,8 Cl A,was rapidly and completely biotransformed into its main metabolite 8 Cl Ad and then 8 Cl Ⅰ 8 Cl A and its metabolites had no accumulative effect in blood.8 Cl A was mainly biotransformed into 8 Cl Ad which distributed in many organs and tissues in dogs.
