Preliminary Study on Apoptosis and Expression of Apoptosisrelated Genes in Peripheral Blood Lymphocytes of Patients with Systemic Lupus Erythematosus

摘  要:为探讨细胞凋亡及其相关基因与系统性红斑狼疮(SLE)患者病情的关系,作者采用末端脱氧核苷酸转移酶介导的缺口末端标记法及流式细胞仪,观察了29例SLE患者外周血淋巴细胞(PBL)凋亡现象,并应用免疫组化ABC法检测了bcl-2蛋白和Fas蛋白的表达。结果显示:①SLE患者PBL凋亡率明显高于正常组及类风湿关节炎组(P<0.001),经48小时培养后PBL凋亡率更高,可达33.44%;②PBL凋亡率与疾病活动程度呈正相关(r=0.86,P<0.001);③并发狼疮肾炎(LN)者,其PBL凋亡率较无LN者高;④培养前后,SLE患者外周血淋巴细胞bcl-2蛋白及Fas蛋白表达均显著高于正常组(分别为P<0.01,P<0.001)。因此,PBL凋亡率的变化可能作为评估SLE患者病情的一种指标。This study was conducted to explore the clinical implication of the relationship between systemic lupus erythematosus (SLE) and peripheral blood lymphocytes (PBL) apoptosis and apoptosisrelated gene expression. Terminal deoxynucleotidyl transferase mediated nick end labelling technique and flow cytometry were used to detect the apoptotic cell; immunohistochemistry ABC was used to detect the expressions of bcl2 and Fas in 29 cases of SLE. The results showed that the rate of apoptosis of PBL in SLE patients was significantly higher than that in normal control (P<0.001), and that in patients with rheumatoid arthritis (P<0.001). After 48 hour's culture, the apoptotic rate further increased in SLE patients (33.44%). There was a significant correlation between the SLE activity and the rate of apoptosis in vitro (r=0.86,P<0001). The rate of apoptosis of PBL in SLEnephritis patients was higher than that in nonSLEnephritis patients (P<001). Before and after the culture, the expressions of bcl2 and Fas in PBL
