摘 要:目的:初步探讨β2ARmRNA在支气管哮喘发病中的变化。方法:用RT-PCR及原位杂交方法测定哮喘时人外周淋巴细胞及大鼠肺组织β2ARmRNA水平。结果:缓解期哮喘患者β2ARmRNA水平与正常人无显著差异。发作期哮喘大鼠肺组织β2ARmRNA水平显著降低,且肺内不同组织细胞β2ARmRNA水平的下降程度并不一致。结论:提示哮喘时β2ARmRNA的变化可能是β2AR变化的分子基础,这对哮喘治疗具有重要的实际意义。AIM:To preliminaryly inquire into the changes of β 2AR mRNA levels in bronchial asthma.METHODS: To measure the β 2AR mRNA levels in asthmatic peripheral lymphocytes and the lungs of challenged rats by using RT-PCR and in situ hybridization.RESULTS:(1) Expression of β 2AR mRNA in peripheral lymphocytes of asthmatics at remission stage was no significant differences compared with that in normal volunteers; (2) Expression of β 2AR mRNA in the lungs of challenged rats decreased markedly, but β 2AR mRNA levels among various tissues inside lung decreased to different extent after allergen challenge.CONCLUSION: Changes of β 2AR mRNA might underlie the changes of β 2AR in the lungs of asthmatics and challenged rats, and it also furnished theoretical basis for the useage of β 2AR agonists clinically.
