摘 要:为了解香猪睾丸发育过程中的形态学变化,外科手术取30、40、50、60、90、110日龄(每个年龄组n=3~4)香猪右侧睾丸经中性多聚甲醛固定,石蜡包埋,组织切片采用HE染色,40倍物镜下观察细胞数量变化,经显微照相后用Scion image软件测量生精小管、输出小管和附睾管的管壁面积。结果发现30~50日龄时睾丸生精小管和管壁面积无显著变化(P>0.05),从60日龄开始到110日龄出现显著性扩大(P<0.05),输出小管、附睾管及其管壁面积均随着日龄增长而增加(P<0.05)。30、40和50日龄时睾丸输出小管和附睾管的细胞数量变化并不显著(P>0.05),而从60日龄开始,睾丸输出小管和附睾管的细胞数量大量增加(P<0.05)。30、40、50日龄睾丸生精小管中只发现有少量生殖细胞,但60~110日龄的睾丸生殖细胞数量快速增多(P<0.05)。生精小管与输出小管管壁单位面积细胞数均随日龄增长而增加(P<0.05),而在附睾管出现下降(P<0.05)。To investigate the morphology in testis development of male xiang pig.30、40、50、60、90and110-day-old pig (number=3~4) right testis were collected by surgical operation.The collected tissues were fixed in 4% (w/v) paraformaldehyde for 24 h and then processed to paraffin wax.Five-micron-thick sections were cut for staining by HE and cell number was observed with micscop.The area of seminiferous and epididymis tubule was measured with Scion image software after micrography.Sertoli cell were studied by immunohistochemistry using a specific monoclonal antibody to the GATA-4.Antibody could recognize its specific protein of sertoli cell and couldn't recognize its specific protein of germ cell in tissues and accounting the number of sertoli cells and germ cells in the testis.The results showed that the area of seminiferous tubule range from(6 409.97±485.38)to(7 873.62±1 359.06)μm2at 30~50day(P0.05).It rose to(23 683.16±4 141.17)μm2 at 60 d and significantly higther than that of 30~50 d(P0.05),then it continuing significantly increasing until 110d(24 879.18±4 193.88)μm2(P0.05).The area of epididymis and efferente tubules were increasing from 30 d until 110 d(P0.05).The number of sertoli cell were not changed from 30 d (32.58±5.74) to 110 d (28.21±3.36) (P0.05).The number of germ cell didn't change at 30~50 d(24.10±2.66)^(26.23±3.36).but they were significantly increasing from 60 d(140.50±23.39) to 110 d(259.60±28.03) (P0.05).
