落叶松体细胞胚胎发生过程中DNA甲基化模式变化分析

摘　　要：通过MSAP技术检测落叶松体细胞胚胎发生过程中的DNA甲基化模式变化发现,体细胞胚胎发生的14个时期中,5’-CCGG-3’序列的胞嘧啶上发生重新甲基化事件最多在ABA诱导后的0.5～6 h,最少在5～7d的早期单胚阶段;而去甲基化事件发生最多的是5～7 d的早期单胚阶段,最少在14～21 d的中期单胚到晚期单胚阶段。与原胚团时期相比,重新甲基化事件最多发生在ABA诱导后的5 d,最少发生在ABA诱导后的7 d;去甲基化事件发生最多是在中期单胚的14 d,最少是在ABA诱导后的2 d。说明落叶松体细胞胚胎发生过程中存在着频繁的重新甲基化和去甲基化变化,体细胞胚胎成熟阶段与原胚团阶段相比,DNA甲基化的模式变化显著。A study was conducted to detect DNA methylation pattern changes during 14 stages of somatic embryogenesis of Larix by methylation sensitive amplification polymorphisms（MSAP） technique.Results showed that de novo methylation happened mostly during 0.5-6.0 h after ABA induction and the least of de novo methylation occurred at 5-7 d during the early period of single embryo.However,most demethylation happened at 5-7 d during the early period of single embryo and the least of demethylation occurred at 14-21 d during morphological transformation from the middle period of single embryo to the late period of single embryo.Compared with proembryo stage,de novo methylation happened mostly at 5 d and seldom at 7 d after ABA induction,while demethylation happened mostly at 14 d during the middle period of single embryo and occurred seldom at 2 d after ABA induction.All of the results confirm that the most frequent changes of methylation pattern exist in the somatic embryogenesis of Larix,and the maturation stage of somatic embryogenesis has significant changes in DNA methylation pattern compared with the proembryo stage.
