摘 要:目的探讨姜黄素衍生物FM0817体外抗淋菌作用及其机制。方法应用K-B法检测40株淋菌对抗菌药物的敏感性,煮沸法提取菌株DNA,随机抽取其中22株PCR扩增TEM、tet M、erm和mef A基因,采用滤纸片扩散法,初步判断FM0817是否有抗菌作用,作用结果结合耐药表型和基因型分析FM0817体外抗淋菌作用可能的机制。结果淋菌的药敏率测定显示多药耐药明显;TEM、tet M的检出率均为77.2%,未检出erm和mef A基因;FM0817抗淋菌作用明显。结论姜黄素衍生物FM0817具有较强的体外抗淋菌作用,有进一步开发研究的前景,其作用机制与淋菌耐药表型和基因型未发现有关系。OBJECTIVE To analyze the antigonococcus activity of curcumin derivative FM0817 and its mechanism. METHODS A total of 40 strains of N. gonorrhoeae were tested their susceptibilities to antibiotics by K-B and agar dilution methods. Total DNA of 22 strains were extracted by boiling methods, which were used as a template in PCR to amplify the drug resistant genes of TEM, terM, erm and mefA. RESULTS The susceptibilities rates showed that the multi-drug resistance of N. gonorrhoeaewas was obvious. The detection rates of TEM explicit and terM were 77.2% and 77.2%, respectively. The genes of erm and mefA were not detected in the study subjects. CONCLUSION FM0817 exhibits potent activity in antigonococcus in vitro and deserves further research and development. It has not found that the activity mechanism is related to the drug resistance of N. gonorrhoeae and the genotype.
