摘 要:目的了解静脉血白细胞过滤模型对犬白细胞免疫功能的影响。方法 21只犬静脉麻醉后随机分为两组。白细胞滤除组11只犬双侧颈外静脉穿刺后行静脉血白细胞过滤。对照组10只犬双侧颈外静脉穿刺后持续镇静。以开始白细胞过滤为起始时间,检测白细胞滤除组犬0、1、2、6 h及对照组相对应时间点TNF-α、IL-6浓度、中性粒细胞上CD11/18、单核细胞上CD14的平均荧光强度。将犬处死后取右上肺组织测定髓质过氧化物酶(MPO)活性及HE染色行镜下病理检查。结果两组血浆TNF-α浓度,中性粒细胞上CD11/18,单核细胞上CD14的表达无统计学差异。白细胞滤除组IL-6浓度在1h点和2h点明显低于对照组。两组肺组织MPO活力无统计学差异。两组肺病理切片均无明显病理学改变。结论静脉血白细胞过滤法不会对机体脏器造成炎性损伤,并能降低部分炎性因子。Objective To find out about the effect of a venous blood leukocyte filtration model on leukocyte immune function in canines. Methods Twenty - one mongrel dogs were anesthetized intravenously and randomized into two groups : the leukocyte filtration group (LF group) and the control group( C group). In the LF group, venous blood leukocytes in dogs were filtrated by aseptic puncture of the bilateral external jugular veins. In the control group, dogs were anesthetized continuously after aseptic puncture of the bilateral external jugular veins. The level of TNF - a and IL -6 and the expression of CD11/18 and CD14 were tested in both LF group and C group at Oh, I h, 2h and 6h. MPO enzyme activity and histopathology in the lung in both groups was measured at 6h. Results There was no significant difference in TNF - c~ level, expression of CD11/18 and CD14, MPO enzyme aetivity between LF and control groups. There was no change of histopathology in the lung between LF and control groups. The IL - 6 concentration was reduced at lh and 2h. Conclusion Venous blood leukocyte filtration will not activate immunofunction of the body or degrade some inflammatory factors.
