肾康灵干预阿霉素肾病大鼠血蛋白质组的实验研究

摘  要:目的应用蛋白质组学筛选阿霉素肾病大鼠血清中差异表达的蛋白质,旨在观察频复发性肾病综合征的诊断标志物及肾康灵的作用靶点。方法利用表面增强激光解析-电离飞行时间质谱合CM10蛋白质芯片技术检测5组大鼠血蛋白质组质谱的表达,用Ciphergen Proteinchip Soft-ware 3.2.1和Biomarker Wizard分析软件筛选出5组大鼠血清差异表达的蛋白质。结果在质荷比值2000~50000范围内,P<0.05,质谱仪筛选出B组与A组比较的25种差异蛋白,C组与B组比较筛选出5种差异蛋白,其中3种差异蛋白在B、C、D、E组间有显著差异,且从B组→C组→D、E组→A组呈递减趋势。结论表面增强激光解析-电离飞行时间质谱能有效分析阿霉素肾病大鼠血清蛋白质质谱,肾康灵干预治疗后蛋白质质谱表达有显著差异,且肾康灵合强的松组治疗阿霉素肾病大鼠的疗效优于强的松组。Objective To screen the serum proteomic patterns of rats with adriamyein-induced nephropathy by serum proteomies for discussion of the biomarker of frequent relapse nephrotic syndrome and target point of medication. Methods Surface enhanced laser desorption/ionization-time of flight-mass spec- trometry (SELDI-TOF-MS) and CM10 protein clips was used to detect the serum proteomie patterns of rats in the five groups, and proteins with differential expression of each group were analyzed by Ciphergen Proteinchip Software 3.2.1 and Biomarker Wizard software. Results Proteins with differential expression were detected in the mass charge ratio range of 2 000-50 000 (P〈0.05), there were 25 proteins between group B and group A, 5 proteins between group B and group C, 3 proteins decreased progressively from the group B, to the group C, to the group D, group E, and to the group A. Conclusion The serum proteomic patterns of rats with adriamycin-indueed nephropathy can be analyzed effectively by SELDI-TOF-MS. The difference of proteomics was significant before and after intervention of Shenkangling. The curative effects of Shenkangling combined with prednisone are superior to those of prednisone alone for the treatment of adriamycin-induced nephrotic rats.
