摘 要:目的探讨补肾中药对帕金森病(PD)模型小鼠黑质-纹状体多巴胺(DA)的影响。方法用MPTP.HCl腹腔注射法建立PD小鼠模型,选择造模成功小鼠随机分为淫羊藿组、黄精组、司来吉兰组、模型对照组,每组6只,另取同周龄同品系小鼠6只作为正常对照组,正常对照组、模型对照组灌胃给予蒸馏水,淫羊藿组、黄精组、司来吉兰组灌胃给予淫羊藿、黄精、司来吉兰的水煎液或水溶液,连续给药4周后透取小鼠脑脊液,HPLC-EC法检测DA及高香草酸(HVA)的含量。结果淫羊藿组、黄精组DA的含量明显高于模型对照组(P<0.05),低于空白对照组(P<0.05)。结论淫羊藿、黄精可增加PD模型小鼠黒质-纹状体中DA的含量。Objective To investigate the effect of Chinese herbs for tonifying kidney on striatal dopamine (DA) content of mouse model of Parkinson's disease (PD). Methods The models were established with peritoneal injection of MPTP. HC1, then the model mice were divided randomly into Epimedium group, Polygonatum group, selegiline group and model group, each group had 6 mice, and 6 same age and strain mice as normal control. Normal control group and model group were given distilled water by gavage. Epimedium group, Polygonatum group and selegiline group were given Polygonatum solution, Polygonatum solution and selegiline solution respectively. After successive administration for 4 weeks, cerebrospinal fluid of mice was separated, and the contents of DA and HVA were detected by HPLC-EC. Results The content of DA in Epimedium group, Polygonatum group was higher significantly than that of model group respectively (P〈0.05), and lower significantly than that of normal control group respectively (P〈0.05). Conclusion Epimedium and Polygonatum can increase the DA content in strbstantia nigra-striatum of PD model mice.
