摘  要:目的:探讨电针对易脑卒中型肾血管性高血压大鼠(RHRSP)脑缺血再灌注不同时点促血管生成素-1(Ang-1)及凝血酶敏感蛋白-1(TSP-1)表达及血管新生的影响。方法:用环形银夹钳夹SD大鼠的双侧肾动脉,制成120只RHRSP,随机分为4组:空白组(12只),假手术组(12只)、模型组(48只)和电针组(48只),后两组大鼠再用线栓法制成一侧大脑中动脉闭塞(MACO)模型,电针组给予"百会"、"大椎"穴位电针治疗,1次/d,14d。观察各时间点大鼠行为学评分及超微结构变化;用免疫组织化学法和电镜等技术观察脑缺血2h后再灌注1、7、14、28d大鼠脑内缺血区Ang-1、TSP-1表达及缺血区微血管密度(MVD)的变化。结果:电针组大鼠行为学评分在1、7、28d明显低于模型组(P<0.05);电针组和模型组脑组织含水量在1、7d均明显高于空白组与假手术组(P<0.05),但模型组脑组织含水量在1、7d较电针组升高更明显(P<0.05);与模型组比较,电针组Ang-1的表达在7、14、28d增强(P<0.05),TSP-1的表达在1、7、14d减少(P<0.05),MVD在14、28d增加(P<0.05)。超微结构检查发现电针组各时间点脑组织神经细胞与血管内皮细胞的损伤较模型组减轻。结论:电针对RHRSP脑缺血-再灌注损伤的保护作用,可能与上调Ang-1及下调TSP-1的表达,促进脑缺血区的血管新生有关。Objective: To investigate the influence of electric acupuncture on the expression of angiopoietin-1(Ang-1),thrombospodin-1(TSP-1) and angiogenesis during the different time of the cerebral ischemia and reperfusion in Stroke-prone-Renovascular-Hyper-tensive-Rats(RHRSP).Methods: 120 Renovascular hypertensive rats with stroke-prone(RHRSP) models were established by bilaterally narrowing the renal artery with sliver loop clips and then randomly divided into four groups: blank group(12),sham-operated group(12),model group(48) and electric acupuncture group(48).The middle cerebral artery occlusion(MCAO) models were duplicated by thread occlusion method in model group and electric acupuncture group.EA at Bai Hui and Da Zhui points were carried out.in electric acupuncture group once a day for 14 days.The behavioral scores and ultrastructural changes of rats were observed at each time point.The expression of Ang-1,TSP-1 and changes of microvessel density(MVD) in ischemic area of rats were detected by immunohistochemistry,electron microscope on 1th,7th,14th,28th days 2h after cerebral ischemia and reperfusion.Results: The behavioral scores of rats in electric acupuncture group decreased significantly than that in model group(P0.05) on 1th,7th,28th days.The brain water content of rats in electric acupuncture group and model group increased obviously than that in blank group and sham-operated group,and it was higher in contral group than that in electric acupuncture group on 1th,7th days.Compared with model group,the expression of Ang-1 was strengthened(P0.05) on 7th,14th,28th days but TSP-1 was lowered(P0.05) on 1th,7th,14th days in electric acupuncture group.The microvessel density in and around ischemic area increased obviously in electric acupuncture group than those in contral group(P0.05) on 14th,28th days.The injury of neurons and endothelial cells in electric acupuncture group was more obviously alleviated than that in model group at each time point.Conc
