摘 要:采用我国流行的CB4V亲胰腺株(P-CB4V)感染Swiss小鼠,建立Ⅰ型糖尿病的动物模型,并观察黄芪对CB4V感染引起-Ⅰ型糖尿病的保护作用。结果表明,80%的小鼠在感染后72h出现低血糖,血糖在(90.87±8.26)mg/L范围之内,6~8周全部发展为高血糖,糖刺激的胰岛素释放量在感染后72h增加,但至3~8周明显低于未感染组。在用VB4V感染小鼠的同时给予黄芪,目的在于观察黄芪对CB4V攻击胰岛β细胞的保护作用,结果经黄芪治疗后的感染小鼠,1周80%小鼠血糖在(112.66±5.0)mg/L,范围之内,60%小鼠低糖及高糖刺激的胰岛素释放量与正常对照组无差异,到3~6周全部小鼠血糖在(113.68±6.30)mg/L,范围之内,而低糖与高糖刺激的胰岛素释放量分别在(20.22±285)uIU/ml,(3640±3.05)uIU/ml范围之内,均与正常对照组无差异(P<0.05)。Swiss mince infected with pancreas-adapted strain of CB4V (P- CB4V), which is epidemicin China, were used to establish and induce animal model of type I diabetes mellitus and observethe protective effect of Astragalus membranaceus on the disease. The results indicated tha hypoglycemia appeared in 80% of the mice at 72h p. i. with blood sugar level at 90. 87±8.26mg/L,and hypergycemia in total mice from 6W to 8W. Insulin secretion level was increased after stimulation of sugar at 72h p. i., however it was decreased from 3W to 8W as compared with controlledgroup. Asragalus memranaceus were given to the mice at the same time of CB4 V infection, toobserve its protection action on β-cell which is the target of CB4V. The results showed thatblood sugar level was at 112. 66±5. 0mg/L in 80% of the therapeutic mice by Astragalus membranaceus in 1W p. i., and insulin secretion level stimulated by hypoglycemia and hyperglycemiain 60% of the mice showed no difference as compared with the oontrolled group. From 3W to6W, the blood sugar level was at 113. 68±6. 30mg/L, while insulin secretdrion stimulated by hypoglycemia and hyperglycemia was at 20. 22 ± 2. 85uIU/ml. And 36. 40±3. 05uIU/ml respectively, which showed no differene with the controlled group (P>0. 05).
