摘 要:目的:观察山茱萸环烯醚萜苷(CIG)对脑缺血动物学习记忆能力以及海马区BDNF蛋白表达的影响。方法:沙土鼠双颈总动脉夹闭阻断血流5min造成全脑缺血模型。4~5周后,通过避暗试验和Morris水迷宫试验观察动物学习记忆能力;应用尼氏染色法检测海马区神经元的损伤程度;免疫组织化学和免疫印迹方法检查脑海马BDNF的表达情况。结果:灌胃给予CIG可以明显改善全脑缺血沙土鼠的被动回避记忆和空间学习谒忆能力,显著提高海马区BDNF的表达,保护CA1区神经元细胞免受缺血性损害。结论:CIG具有神经保护作用,其机制可能与促进海马区神经营养因子的表达有关。Objective: To investigate the effects of Cornel iridoid glycoside (CIG)on the learning and memory ability and neurotrophic factors in the brain of global ischemic gerbils. Methods :The global brain ischemic model was made by occluding common carotid arteries for 5 min in gerbils. After 4 to 5 weeks, passive avoidance memory and spatial learning - memory abilities were performed by step through and Morris water maze tests, respectively. Nissl staining was used to observe the survival neurons in hippocampal CA1 area. The expression of brain - derived neurotrophic factors (BDNF) was detected with immunohistochemical staining and western blotting. Results : Intragastrical administration of CIG significantly improved the passive avoidance memory and spatial learning - memory abilities, increased the expression of BDNF in hippocampal CA1 area and protected CA1 neurons against ischemia damage in gerbils. Conclusions :GIG can improve the cognitive function of gerbils suffered from global brain ischemia. The mechanism may involve increasing the expression of BDNF, thereby protecting CA1 neurons against ischemie insults.
