摘 要:目的:观察超微粉化瓜蒌根对2型糖尿病(T2DM)SD大鼠免疫机制异常的干预研究。方法:选用雄性SD大鼠,予高脂高盐饲料喂养8周后加小剂量链脲佐菌素(STZ)注射建立T2DM大鼠模型,大鼠随机分为正常对照组,模型组,中药(低、中、高剂量)组。中药组分别予微粉化瓜蒌根灌胃,正常组和模型组给予等体积生理盐水,治疗4周后采用稳态模式法(HOMA)评价胰岛素抵抗(Homa-IR)和胰岛β细胞分泌功能(Homa-IS)、检测空腹血糖(FPG)及空腹胰岛素(Fins);应用流式细胞仪检测血淋巴细胞亚群分布的变化情况(CD系列)。结果:T2DM大鼠模型呈现CD4+T细胞、CD19+B细胞、CD11a+表达水平显著低下(P<0.05);中药治疗后FPG、Ho-ma-IR显著低于模型组(P<0.05),Homa-IS则显著高于模型组(P<0.05);中药组的CD4+表达显著低于模型组、CD56+、CD14+的表达显著高于模型组(P<0.05)。结论:T2DM大鼠模型存在着特异、非特异免疫细胞功能低下;超微粉化瓜蒌根通过调节淋巴细胞亚群比例分布及降糖、改善胰岛素抵抗和胰岛β细胞分泌功能,对T2DM大鼠具有免疫调节作用。Objective : To observe the effect of uhrafine powder of the root of gualou on abnormal immune response in type 2 diabetic SD rats. Methods: Type 2 diabetic mellitus (T2DM) was induced in male SD rats by giving high -sugar, high - salt forage for 8 weeks and then injecting low - dose streptozotocin. Rats were randomly divided into normal control group, model group, and gualou groups (low - , medium - , and high - dose subgroups). Gualou was given intragasti- cally to the gualou groups and saline to the normal control and model group in an equivalent volume for 4 weeks. Homa - IR, Homa - IS, fasting plasma glucose and insulin were detected. Lymphocyte subsets were determined by flow cytometry. Results: T2DM rats had CD4^+ T cells, CD19^+ B cells, and CD11a^+ cells significantly decreased( P 〈 0.05 ). The levels of fasting plasma glucose and Homa - IR were reduced in the gualou groups as compared to the model group ( P 〈 0.05). CD4^+ T cells was decreased but CD56^+ and CD14^+ were increased in the gualou groups as compared to the model group( P 〈0.05). Conclusion: T2DM rats have abnormal immune function. Uhrafine powder of the root of gualou may regulate the immune function in T2DM by influencing the lymphocyte subset distribution, decreasing plasma glucose, and improving insulin resistance and 13 cell function.
