摘 要:目的:研究补阳还五汤对局灶性脑缺血大鼠血管生成和神经功能恢复的影响。方法:采用线栓法诱导大鼠大脑中动脉阻塞,缺血90min后再灌,缺血后24h开始灌胃补阳还五汤,1天1次,连续14天,在缺血后第1、3、7、14天分别采用肢体放置实验、错步实验和粘胶揭除实验评价感觉运动功能;采用免疫组化检测缺血周边区微血管密度(microvessels density,MVD)和血管内皮生长因子(vascular endothelial growth factor,VEGF)表达。结果:与模型组比较,补阳还五汤不仅能显著提高肢体放置实验成绩,减少错步次数和缩短粘胶揭除潜伏期(P<0.05或P<0.01),而且能显著增加缺血周边区vWF阳性血管和VEGF阳性细胞数量(P<0.01)。结论:补阳还五汤能促进脑缺血后神经功能恢复,机制可能与其促进缺血周边区血管生成和VEGF表达有关。Objective : To investigate the effect of Buyanghuanwu decoction ( BYHWD ) on angiogenesis and functional recovery after focal cerebral ischemia in rats. Methods : Focal cerebral ischemia was induced by 90 min of middle cerebral artery occlusion followed by reperfusion for 14 days. BYHWD(13g/kg) was administered orally 24 h after ischemia and once a day for 14 days. The limb placing test, the foot fault test, and the adhesive tape test were used to evaluate sensori- motor function during the 1, 3, 7 and 14 days after ischemia. Thereafter, micrevessels density(MVD) and vascular endothehal growth factor(VEGF) expression in the ischemic boundary were determined by immunohistochemistry. Results: Compared with the model group, BYHWD not only improved the score of limb placing, reduced the number of foot fault and latency time of removal adhesive tape, but also significantly increased the numbers of VWF positive microvessels and VEGF positive cells at the border of the infarct. Conclusion : BYHWD improved functional recovery after focal cerebral ischemia, which may be associated with the enhanced angiogenesis and VEGF expression in the ischemic boundary.
