血府逐瘀汤促急性心肌缺血大鼠心肌血管新生及对血管内皮细胞生长因子的影响

摘　　要：目的探讨血府逐瘀汤对急性心肌缺血大鼠缺血心肌血管新生的影响及其分子学机制。方法雄性SD大鼠,结扎冠状动脉左前降支复制急性心肌缺血模型,随机分为模型组、血府逐瘀汤组、麝香保心丸组,并设假手术组。造模后24 h各组灌胃给予相应药物或蒸馏水,每日1次,连续7 d,于第7日灌胃后1 h断头处死动物,取出心脏。用免疫组化法测定缺血心肌内皮细胞数和微血管密度,检测缺血心肌局部血管内皮细胞生长因子(VEGF)的表达。结果血府逐瘀汤组内皮细胞数、微血管密度及VEGF表达均明显升高,与模型组比较,差异有统计学意义(P<0.05)。结论血府逐瘀汤能够促进心肌缺血大鼠缺血心肌血管新生,对缺血心肌具有保护作用,其作用机制可能与上调心肌局部VEGF蛋白表达有关。Objective To explore the protective effects of Xuefu Zhuyu Decoction of myocardial angiogenesis and molecular mechanisms rats with acute myocardial isehemia. Methods The left anterior descending coronary artery was ligated to make the model of myocardial ischemia in SD rats. The rats were divided into 4 groups at random： the model group, Shexiangbaoxin pills group, Xuefu Zhuyu Decoction group and the sham operation group, with 8 rats in each group. All groups were treated for 7 days by corresponding drug. After 7 days, all rats were killed and the hearts were taken for detecting cardiac endothelial cell and the microvessel vascular density （MVD）. The expression of VEGF at protein level was studied by immuneohistoehemical method. Results Compared with model group, Xuefu Zhuyu Decoction could significantly enhance the levels of MVD and expression of VEGF （P〈0.05）. Conclusion Xuefu Zhuyu Decoction can promote angiogenesis in ischemic myocardium of rats. It has protective effect on acute myocardial ischemia in rats. Upregulating the expression of VEGF may be one of its mechanisms.
