摘  要:目的在重组人骨形态发生蛋白-2(rhBMP-2)诱导异位成骨过程中,局部应用不同浓度的辛伐他汀(SIM),探讨SIM对rhBMP-2诱导成骨的作用。方法①以牛松质骨为原料经过脱脂、脱蛋白等步骤制备牛骨基质明胶载体,将辛伐他汀和rhBMP-2分别与BMG载体复合;②将45只雄性SD大鼠随机分为3组。A组:高剂量实验组;B组:低剂量实验组;C组:对照组;③将复合体植入大鼠股后部肌袋中,术后15d、30d、60d对实验大鼠进行大体观察、X线检测。结果①大体观察:各组组织块均随着时间延长而变硬,范围变大;②X线结果,各组植入部位术后15d、30d未见显影,于术后60d各组可见不同密度的影像,B组和C组钙化明显,成高密度影像;A组成低密度影像。成骨区灰度值A组明显低于B组和C组。结论高浓度辛伐他汀可明显抑制rhBMP-2的促进成骨作用;低浓度辛伐他汀的抑制作用则不明显;证实了rhBMP-2在促进成骨方面有重要作用。Objective To investigate the synergistic effect of different concentration Simvastat(SIM)on the bone induction of recombinant human bone morphogenetic protein-2(rhBMP-2)through local application of SIM in the osteoinductive process of BMP.Methods ①Prepare the bone matrix galatin(BMG)carrier as the raw materiar from cattle spongy bone by the procedure of degrease, deproteinization and so on.To compound the BMG carrier with SIM、rhBMP-2 or both.②Forty-five male SD rat were divided into two experimental groups and control group at random,and BMG composite was implanted into the right thigh muscle pouch of each group. group A:the BMG carrier contain 15% weight ratio of SIM and 0.25 mgrhBMP-2 as high dose experimental group;group B:the BMG carrier contain 2.5% weight ratio of SIM and 0.25 mgrhBMP-2 as low dose experimental group; group C:the BMG carrier just contain 0.25 mgrhBMP-2 as control group(CG). ③At dl5, d30 and d60 after implantation, new bone formation was measured by generally, radiographically of the three groups.Results ①The carriers of each group become hard and extensive. The effect of wet weight:the groups of A, B, C were different, which the group C has the strongest effection of all. ②X-ray results:Each group can be seen the developing after 60 days from operation, which group B and Group C have obviously calcification.The detection of gray scale on the zone of ossification:group B and Group C are much higher than group A.Conclusion The Simvastat of high concentration group can inhibit the osteoogenesis of rhBMP-2 obviously,however,the low concentration group have not obviously inhibitory action.Confirm the important effectiveness of rhBMP-2 on the osteoinductive process.
