摘 要:目的观察补肾化痰法对AD模型大鼠海马皮质区胆碱能系统的影响并探讨其可能机制。方法应用脑立体定向技术给大鼠BNM注射凝聚态Aβ25~35+IBO构建AD模型。注射2周后,高、中、低剂量组分别给大鼠该药水煎液灌胃;西药组用哈伯因混悬液灌胃;正常组、空白组、模型组均给予等容积的生理盐水灌胃。28天后采用放射免疫法测定大鼠海马区Ach、AchE和ChAT活性。结果模型组海马区Ach和ChAT与正常组相比均显著减少(P<0.01),AchE活性则明显升高(P<0.01)。Ach结果正常组与空白组无明显差异(P>0.05)。西药组Ach与中、低剂量组比较有差异(P<0.01),但与高剂量组比较无差异(P>0.05)。从ChAT和AchE活性检测结果看,两者在各组比较趋势一致,各治疗组与模型组比较均有显著差异(P<0.01),低剂量组与西药组比较有差异(P<0.05),与中、高剂量组比较无差异(P>0.05)。各中、高剂量组比较也无明显差异(P>0.05)。结论补肾化痰法对中枢胆碱能损伤具有保护作用,从而发挥其抗老年性痴呆的作用。Objective to explore the effect and potential mechanism of rat hippocampal cortex cholinergic system on replenishing kidney-essence and removing phlegm therapy.Methods To set up AD model,five groups rats received injection of IBO and Aβ25-35 with basal nucleus of Meynert(BNM) through stereospeetific instrument.After treating for two weeks,BuShen HuaTan decoction were administered by oral gavage for the three doses groups.Huperzine A-Zhulin Antun tablets served as control drug.Normal group,model group and blank group were administered by isovolumetric saline water.After treating for four weeks,radioimmunoassay was used to detect the activity of Rathippocampus Ach,AchE and ChAT.Results The expression of Ach and ChAT in model group have marked reduced than normal group(P 0.01),but the activity of AchE increased significantly(P 0.01).The expression of Ach in normal group and group no obvious difference(P 0.05).Ach in western medicine group and the difference between dose groups and low dose groups(P 0.01),But no difference between high dose group(P 0.05).From the measured results of ChAT and AchE activity,the treatment group and model group were significantly different(P 0.01),low-dose group compared with the western medicine group differences(P 0.05),And no difference in the dose group、high-dose group,(P 0.05 ).The medium dose group,high-dose group showed no significant difference(P 0.05 ).Conclusion By replenishing kidney-essence and removing phlegm therapy has a protective effect on the Cholinergic damage,to play the role of anti-Alzheimer.
