摘  要:目的:应用近红外光谱法和数据分析软件,对金银花中绿原酸含量进行快速测定。方法:利用HPLC测定样品中绿原酸的含量,运用偏最小二乘(PLS)法建立其含量与NIR光谱之间的多元校正模型,对未知样品进行含量预测。结果:建立的绿原酸校正模型相关系数(R2)、内部交叉验证均方差(RMSECV)、校正均方差(RMSEC)分别为0.993,0.380,0.158。经外部验证,校正模型的预测均方差(RMSEP)、平均回收率分别为0.170%,101.78%。结论:此方法具有快速简便,准确无损的特点,可以应用于金银花中绿原酸含量的快速检测。Objective:To determine chlorogenic acid of honeysuckle rapidly by near-infrared spectroscopy and data analysis software. Method: HPLC was used as a reference method to determine the content Of chlorogenic acid in honeysuckle. Multivariate calibration model based 'on PLS algorithm wasdeveloped to correlate the spectra and the corresponding values determined by the reference method. Result:The correlation coefficients(R2) ,the rootmean-square error of cross-validation (RMSECV) and the root-mean-square error of calibration(RMSEC) of the calibration model for chlorogenic acid were 0. 993,0. 380 and 0. 158, respectively; the root-mean-square error of prediction(RMSEP) and the average rate of recovery were 0. 170 and 101.78%. Conclusion:The method is fast,convenient,accurate and lossless. The correction model could be used to predict chlorogenic acid of honeysuckle rapidly.
