摘 要:目的:观察熊果酸对大鼠胰岛素抵抗糖代谢的改善及可能作用机制。方法:采用高脂饲料喂养的方法诱导胰岛素抵抗模型。检测大鼠空腹血糖、空腹胰岛素、口服葡萄糖耐量、肝脏葡萄糖激酶含量,葡萄糖激酶mRNA表达水平,葡萄糖激酶蛋白表达。计算胰岛素抵抗指数和胰岛素敏感指数。结果:熊果酸300mg/(kg.d)组和熊果酸150mg/(kg.d)组空腹胰岛素和胰岛素抵抗指数降低,胰岛素敏感指数升高,葡萄糖耐量明显改善,肝脏葡萄糖激酶含量、mRNA及蛋白表达增加。结论:熊果酸对大鼠胰岛素抵抗的改善可能与肝脏葡萄糖激酶表达升高有关。Objective: To observe the effect of ursolic acid on insulin resistance and possible mechanisms. Methods: The insulin resistance model were made by high fat diet. Detect the fasting blood glucose ( FBG), fasting serum insulin ( FIN), the oral glucose tolerance test (OGTI?) , the content of the liver glucokinase, the expression of mRNA and protein of glucokinase. Calculate insulin sensitivity index (ISI) and insulin resistance index (HOMA- IR). Results: In ursolic acid 300rag/ (kg d) group and ursolic acid 150mg/ (kg d) group, the results of FINS, HOMA -IR were decreased, ISI was incresed, and OGTY was improved. The levels of glucokinase content, mRNA and protein expression in the liver were elevated obviously. Conclusion: It is suggested that the therapeutic effects of ursolic acid on insulin resistance might be related to the glucokinase expression in the liver of insulin resistance rats.
