摘  要:目的:观察大鼠口服清热解毒方后,其有效成分大黄酸在大鼠血浆中的经时变化规律。方法:采用RP-HPLC法,色谱柱Kromasil C18(250mm×4.6mm,5μm),以甲醇-0.1%磷酸(85∶15)为流动相,流速1.0mL/min,检测波长254nm,柱温35℃。结果:大鼠口服清热解毒方后,血浆中大黄酸1h达峰值,之后缓慢下降,于12h内基本代谢完全。结论:采用反相高效液相色谱法检测大鼠体内药物含量简便、快捷,适合于研究中药类成分的代谢特征。Objective To observe the ongoing changes of plasma rhein level in mouse after orally taken QingReJieDu (清热解毒)decoction. Methods The RP-HPLC analysis was carried out with a Kromasil C18 (250 mm×4.6 mm, 5 μm) column and a mobile phase of methanol-0.1% phosphoric acid solution (V:V=85:15). The flow rate was 1.0 mL/min and the column temperature was 35 ℃, The detection wave length was set at 254 nm. Results One hour after orally taken QingReJieDu decoction in mouse, the plasma rhein level reached a peak value, then decreased steadily and it was metabolized entirely in 12 hours. Conclusion The ongoing changes of the content of rhein in mouse plasma can be shown by giving QingReJieDu decoction orally. This RP-HPLC is easy and fast to test the body content of some drugs, hence is suitably used in the study of metabolism features of some herbal medicine.
