摘  要:目的:研究雷米普利(Ramipril,RAM)对自发性高血压大鼠(Spontaneously hypertensive rats,SHR)心肌纤维化时心脏瞬时感受器阳离子通道C3亚族表达的影响。方法:12周龄SHR大鼠随机分为安慰剂组、RAM组,同龄WistarKyotorats(WKY)为正常对照组,RAM给药4周后测左室质量指数、心肌胶原容积分数和心脏组织中瞬时感受器阳离子通道C 3(Canonical transient potential receptor channel3,TRPC 3)的mRNA的表达和蛋白的表达。结果:安慰剂组左室质量指数,Ⅰ、Ⅲ型胶原容积分数显著高于WKY组;RAM组上述指标较安慰剂组明显降低。3组心脏均有TRPC3的表达,但安慰剂组TRPC3 mRNA及蛋白表达显著高于WKY组,RAM组TRPC3表达则明显少于SHR组(P<0.05)。结论S:HR及WKY大鼠存在TRPC3蛋白的表达,心脏TRPC3蛋白可能参与了心肌纤维化发生发展的过程,RAM可能通过下调心脏TRPC3 mRNA及蛋白的表达发挥抗心肌纤维化的作用。Objective:To investigate the effect of ramipril on the expression of TRPC3 in the overload induced cardiac fibrosis in the left ventricle of spontaneously hypertensive rats(SHR).Methods:12-week-old SHRs were divided into two groups:the placebo group(SHRs) and the Ramipril group(Ram-SHR),the same aged Wistar Kyoto rats(WKYs) were used as control group(WKY).Four weeks after drug intervention,the left ventricle mass index(LVMI),the cardiac collagen volume fraction(CVF) and the expression of TRPC3 mRNA and protein were detected.Results:The expression of TRPC3 mRNA and protein were detected in each group.Compared with the WKYs,the LVMI,CVF) and the expression of TRPC3 mRNA and protein were significantly increased in SHRs.These data mentioned above were decreased obviously in Ramipril group.Conclusion:Expression of TRPC3 mRNA and protein TRPC3 were found in both SHRs and WKYs.TRPC3 could have play an important role in the development of cardiac fibrosis.Rmipril could inhibit the cardiac fibrosis through down-regulating the expression of TRPC3 mRNA and protein.
