摘  要:目的:研究三聚氰胺在SD大鼠胚胎发育过程中对胎鼠肾的致病性。方法:将40只SD孕鼠随机分为4个三聚氰胺组和1个对照组,用浓度分别为360、480、600、720 mg/(kg.d)三聚氰胺悬液连续灌胃,对照组灌胃2 ml蒸馏水。生产后采用ELISA法检测新生鼠血液和肾脏中三聚氰胺含量,常规HE染色观察新生鼠肾组织的病理变化。结果:各组新生鼠血液中均可检测到三聚氰胺;600、720mg/(kg.d)组新生鼠肾三聚氰胺浓度显著高于对照组。病理组织学检查发现600 mg/(kg.d)组肾小球细胞数量增多,体积增大,髓旁毛细血管扩张充血。720 mg/(kg.d)组肾小管和肾盂扩张,未见结石和明显黄色沉淀物。结论:三聚氰胺通过胎盘屏障进入胎鼠体内,沉积在肾脏,对胎鼠肾生长发育具有损伤作用,损伤主要集中在肾小管和肾间质。Objective:To study the pathogenicity of Melamine in the embryonic kidney development of SD rat.Methods:40 pregnant SD rats were randomized into 4 Melamine groups and the normal control group with 8 rats per group.The rats of the 4 Melamine groups were fed the Melamine with the dosage range of 360,480,600,and 720 mg/(kg·d),respectively.The rats of the normal control group were given 2 ml of distilled water by oral gavage per day.The concentration of Melamine was detected by enzyme-linked immunosorbent assay(ELISA) in the blood and kidneys of the newborn rats.HE staining was used to examine the histopathological change of newborn rat kidneys.Results:Melamine was found in the blood of newborn rats per group.The concentration of melamine in 600,720 mg/(kg·d) dose groups were significantly higher than that of the normal control group(P0.05).Pathological examination showed that hyperplasia of glomerulus in the 600 mg/(kg·d) dose group,and size enlarged,The blood capillary in the renal medulla expanded with RBCs.Kidney tubular and pelvis expanded significantly.The urinary calculus and yellow precipitates were not found in kidneys of 720 mg/(kg·d) dose group.Tubules clearly expanded.Conclusion:Melamine could penetrate into fetal rats through placental barrier,and accumulate in the kidney,which may result in the injury of the growth and development of fetal rat kidney.
