摘 要:为研究Th1型细胞因子IFN-γ、TNF-α在流产发生机制中的意义,探讨中药单体成分川楝素的致流产作用及机理,给妊娠第5天的小鼠连续3d腹腔注射不同剂量的川楝素溶液,对照组以等量的100mL/L丙二醇生理盐水代替,于妊娠第8天剖杀,用ELISA方法检测血清和子宫组织中IFN-γ和TNF-α的水平。结果显示,川楝素的致流产作用呈剂量依赖性,随着注射剂量的增加,小鼠的流产率逐渐上升。随着注射剂量的增加,小鼠血清和子宫组织中IFN-γ、TNF-α的表达水平显著升高,与对照组差异显著(P<0.05或P<0.01)。结果表明,川楝素诱导孕鼠流产与Th1型细胞因子IFN-γ、TNF-α含量的增加有密切关系。In order to study the abortifacient effect and mechanisms of the Chinese herbal medicine component toosendanin,and to elucidate the significance of the Th1 cytokines IFN-γ and TNF-α in the occurrence of abortion,graded doses of toosendanin were given to mice by intraperitoneal on days 5,6 and 7 of gestation.The same volume of 100 mL/L propanediol-normal saline was given to mice in the control group.The levels of IFN-γ and TNF-α in serum and uterine tissues of the mice on day 8 post gestation were analyzed by enzyme linked immunosorbent assay.In result,toosendanin could produce a dose-dependent toxic effect leading to abortion of the mice.The levels of IFN-γ and TNF-α in serum and uterine tissues were significantly increased compare to the control group(P0.05 or P 0.01).The result indicated that toosendanin is toxic to pregnant animals and it is relevant to the increased contents of IFN-γ and TNF-α.
