摘  要:目的研究蛋白激酶C(PKC)3种同工酶PKC α、βⅡ和δ在正常大鼠出生后视网膜发育过程中的表达及意义。方法采用实时荧光定量逆转录-聚合酶链反应(RT-PCR)方法及免疫组织化学方法检测正常大鼠视网膜发育过程中PKC α、βⅡ、δ mRNA及蛋白的表达水平。结果大鼠出生后至第14天,视网膜组织中PKC α、βⅡmRNA表达逐渐升高,14 d后有所下降;PKC δ mRNA在生后早期高表达;视网膜切片免疫组织化学染色光密度测定结果与mRNA表达相符。细胞定位显示,随着视网膜的发育,PKC α染色阳性产物逐渐集中在外丛状层和内核层;PKC βⅡ的阳性产物逐渐表达于视网膜神经节细胞层(RGCs);PKC δ的阳性产物在早期高表达,主要分布在外界膜。结论 PKC α、βⅡ和δ在大鼠出生后视网膜发育过程中均有表达,并有一定的规律性,提示这3种同工酶在大鼠出生后视网膜的发育中起重要作用。Objective To investigate the expression and distribution of protein kinase C α,βⅡ,δ in the retina during the development of postnatal hamster.Methods RT-PCR method was used to detect the expression of mRNA;Immunocytochemical method was used to detect the protein.Results The result of RT-PCR showed that the expression of PKC α,βⅡ mRNA was gradually increased before 14 day of postnatal development and then reduced to the lowest point in the adulthood.PKC δ was highly expressed in the early period of postnatal development.The result of spectrodensitometry matched the result of RT-PCR.With the development of the retina,PKC α was gradually pronounced in the external plexiform layer and inner nuclear layer;PKC βⅡ was pronounced in the ganglion cell layer;In the early period PKC δ was pronounced in the external limiting membrane.Conclusion Protein kinase C α,βⅡ and δ are expressed regularly in the whole development process of rat retina,which suggests that the three isoenzymes are closely related to the early development of retina in the postnatal hamster.
