摘  要:目的观察氯化钴对急性下肢缺血模型缺氧诱导因子-1α(HIF-1α)表达的影响,探讨氯化钴治疗肢体动脉闭塞症的可能性。方法 20只雄性SD大鼠,随机均分成4组:手术+氯化钴、手术+生理盐水、假手术+氯化钴、假手术+生理盐水。手术行左股动脉结扎,术后1 d腹腔注射氯化钴(60 mg/kg),或等容积生理盐水。各组术前1 d、术后1 d(腹腔注射前)、2、4、6 d左腓肠肌穿刺活检。免疫组织化学方法检测HIF-1α的表达。结果股动脉结扎手术可增加术侧肢体骨骼肌HIF-1α的表达(t=5.963,P=0.000),缺血肢体经氯化钴处理后HIF-1α的表达明显高于对照组(F=18.074,P<0.05)。结论氯化钴可以增加缺血肢体骨骼肌的HIF-1α的表达,可望用于治疗肢体动脉闭塞症。Objective To study the effect of CoCl2 on Hypoxia-Inducible Factor-1α(HIF-1α) expression in a hindlimb ischemia rat model.Methods 20 male SD rats were randomly divided into 4 groups(n=5):operation+ CoCl2 group,operation+NS group,sham operation+ CoCl2 group and sham operation+ NS group.Hindlimb ischemia rat model was established by ligating femoral artery.On post operative 1d,all rats received intraperitoneal injection of CoCl2(60 mg/kg) or NS as placebo.The expression of HIF-1α in different groups was detected 1 day before operation and on post operative 1,2,4 d and 6 d respectively by immunohistochemistry.Results The HIF-1α expression in operation groups was higher than that in sham operation groups(t=5.963,P=0.000).And the HIF-1α expression in operation+ CoCl2 groups was also higher than that in operation+NS groups(F=18.074,P0.05).Conclusions Intraperitoneal injection of CoCl2 can increase HIF-1α expression in ischemia limbs.
