摘  要:目的探讨胎儿骨髓间充质干细胞(fBMMSCs)经慢病毒转染胶质细胞源性神经营养因子(GDNF)基因后GDNF的表达情况。方法利用贴壁培养法,从流产胎儿的股骨骨髓中分离培养fBMMSCs,并进行扩增。取第5代fBMMSCs,用慢病毒载体转染GDNF基因。应用免疫组化法及ELISA法检测被转染的fBMMSCs GDNF的表达情况。结果被转染GDNF基因的fBMMSCs GDNF表达阳性,且培养液中GDNF含量随培养时间延长而增高。结论被慢病毒转染GDNF基因的fBMMSCs能表达GDNF。Objective To study on glial cell-derived neurotrophic factor(GDNF) expression of human fetal bone marrow derived mesenchymal stem cells(fBMMSCs) transduced with GDNF gene by lentivirus.Methods The fBMMSCs were isolated from the femur bone marrow of abortive fetus using adherence method and then were cultured and expanded.The GDNF gene was transduced to fifth passage fBMMSCs by lentivirus vector.The expression of GDNF protein in fBMMSCs was detected by immunohistochemical method and ELISA assays.Results The expression of GDNF protein was positive in fBMMSCs transduced GDNF gene,and the content was increased as time prolonger in cultured medium.Conclusion The fBMMSCs transduced GDNF gene by lentivirus can express GDNF.
