摘 要:目的探讨黄芪多糖(APS)对慢性阻塞性肺病(COPD)模型鼠肺组织中血红素氧合酶(HO)1表达的影响。方法 60只大鼠随机均分成5组,每组12只:A组为COPD模型,B1、B2、B3组分别用APS 10、20、40 mg/kg灌胃;另设C组为正常对照。采用香烟熏吸加气管内注入脂多糖(LPS)法建立大鼠COPD模型。RT-PCR与Western blot分别检测大鼠在APS干预前后肺组织HO-1 mRNA及蛋白的表达水平。结果给药后1、15、30 d,A组大鼠肺组织HO-1 mRNA及蛋白水平高于C组(P<0.05)。APS干预30 d后,B3组HO-1 mRNA及蛋白水平较A组明显增加(P<0.05)。结论在COPD模型鼠肺组织中HO-1表达增加;APS可以通过HO-1介导的抗氧化作用来改善COPD大鼠肺组织损伤程度。Objective:To investigate the effects of astragalus polysaccharide(APS) on the expression of heme oxygenase 1(HO-1) in the lung tissue of rats with chronic obstructive pulmonary disease(COPD).Methods:Sixty rats were randomly divided into 5 groups of A(COPD model),B1(treated with 10 mg/kg),B2(treated with 20 mg/kg),B3(treated with 40 mg/kg) and C (normal control).COPD rat model was established by smoke inhalations and intratracheal instillation of lipopolysaccharide(LPS).The expression levels of HO-1 mRNA and protein in the lung tissues were respectively analyzed by RT-PCR and Western blot before and after APS intervention.Results:The expression levels of HO-1 mRNA and protein in group A were higher than those in group C after 1,15,30 d of APS administration(P0.05).After 30 d,HO-1 mRNA and protein levels in group B3 were significantly higher than those in group A(P0.05).Conclusion:HO-1 expression increases in the lung tissue of COPD rat model.APS can reduce lung injury by HO-1 antioxidant.
