两种受孕方式母血、脐血、胎盘中IGF-1的表达

摘　　要：目的检测体外授精-胚胎移植受孕与自然受孕母血、哜血和胎盘组织中胰岛素样生长因子-1（IGF—1）的表达，探讨其差异性及意义。方法收集2007年01月～2008年12月体外受精-胚胎移植受孕行足月剖宫产的产妇49例为研究组，同期因社会因素行剖宫产分娩的正常产妇49例为对照组。采用酶联免疫吸附实验测定两组产妇母血清及脐血清中胰岛素样生长因子-1的浓度；RT—PCR检测两者胎盘组织中胰岛素样生长因子-1mRNA表达水平。采用免疫组化对两者胎盘组织中胰岛素样生长因子-1蛋白进行定位及含量测定。结果与对照组相比，体外授精-胚胎移植受孕组母血清中胰岛素样生长因子-1的浓度略降低，但无统计学差异（t=0．138，P〉0．05），而在脐血清中的表达明显低于自然受孕组，有统计学差异（t=2．556，P〈0．05）；胎盘中胰岛素样生长因子-1mRNA和蛋白表达水平辅助生育组均低于自然受孕组，均有统计学差异（t分别为4．102，4．635，P〈0．05）。结论体外授精-胚胎移植孕妇脐血和胎盘中胰岛素样生长因子-1表达水平较自然受孕组均下降。Objective To detect expression of Insulin-like growth factor-I （ IGF-1 ） in serum, cord blood and placenta of pregnant women following in vitro fertilization and embryo transfer （IVF-ET） and those conceived spontaneously. Methods Serum, cord blood and placenta of 49 term pregnant women after IVF-ET who received cesarean section for term pregnancy （case group） in a period from January, 2007 to December, 2008 and 49 normal term pregnant women who received cesarean section due to social factors （ control grnup） in the same period were taken. IGF-I levels in serum and cord blood of pregnant women in the two groups were determined by enzyme linked immunosorbent assay （ELISA） , and expressinn of IGF-1 mRNA in placenta of pregnant women in the two groups was detected by reverse transeription-polymerase ebain reaction （RT-PCR）. IGF-I protein was orientated and detected by using immunohistochemistry. Results IGF-llevel in maternal serum of pregnant women in ease group was slightly lower than that of the control group, but the difference was not significant（t =0. 138, P 〉0.05）. While IGF-llevel in cord blood in the case group was significantly lower than that in the control group （ t = 2. 556, P 〈 0.05 ） . The expression levels of IGF-1 mRNA and IGF-1 protein in placenta in the case group were both lower than those in the control group （t = 4. 102,4. 635 respectively, both P 〈 0.05 ）. Conclusion The expression levels of IGF-I in cord blood and plaeenta in the case group are both lower than those of pregnant women in the control group.
