摘 要:目的:研究栽培西陵知母中菝葜皂苷元的积累变化规律。方法:采用高效液相蒸发光散射检测,测定不同采集时间知母药材中菝葜皂苷元的含量。色谱柱:Kromasil C18柱(250mm×4.6mm,5μm)。流动相:甲醇,流速1mL.min-1。柱温为30℃。蒸发光散射检测器漂移管温度为50℃,蒸发温度为70℃,以氮气为雾化气,压力为1.03×105Pa。采用胆固醇为内标物,内标法定量。结果:栽培西陵知母各月菝葜皂苷元含量变化较大,尤以6月为最高,7月最低。春季各月含量总体平均高于6月份以后各月。结论:采收期对西陵知母药材质量有较大影响。栽培西陵知母中菝葜皂苷元的含量春季相对较高,春季知母返青前采挖,药材质量较高。Objective:Study on dynamic variation of sarsasapongenin content in cultivated Xiling Zhimu(Anemarrhena Rhizome)from Yi County to investigate its quality.Method:HPLC-ELSD method was adopted to determine the content of sarsasapongenin.With cholesterol as internal standard,Kromasil C18 column(250mm×4.6mm,5μm)was used.Mobile phase was methanol with the flow rate of 1mL·min-1;the temperature of the drift tube and evaporation was 50℃ and 70℃ respectively.The gas pressure was 1.03×105 Pa.Results:The sarsasapongenin content in samples collected in different month varied greatly.Sample collected in June has the highest content,while sample collected in July has the lowest.Samples of spring have a higher average content than those collected after June.Conclusion:The collection time has a great effect on quality of crude drug.Contents of sarsasapongenin in cultivated Anemarrhena Rhizome from Yi County are relative higher in spring.The best harvesting time is early spring.
