摘 要:目的应用化学发光免疫法(CLIA)检测无偿献血者丙型肝炎病毒(HCV)抗体。并与常规抗-HCV ELISA法和HCV RNA-RT-PCR荧光定量检测法进行比较。方法对2009年1月-2010年8月的5 985例献血者血清标本,先应用HCV-CLIA法和试剂①、试剂②两种抗-HCV ELISA试剂进行常规初检、复检检测。对HCV-CLIA法和试剂①、试剂②检出的阳性标本,再进行HCV RNA-RT-PCR荧光定量检测。结果在被检测的5 985份标本中,HCV-CLIA法HCV阳性14份;抗-HCV ELISA法初检阳性15份和复检阳性为16份(其中初检、复检均阳性的14份);HCV RNA-RT-PCR荧光定量检测阳性的13份。结论本组HCV-CLIA法与HCV ELISA法初检、复检结果比较符合率分别为93.33%和87.50%;与HCV RNA-RT-PCR荧光定量检测比较符合率为92.86%。HCV-CLIA法具有操作简便、快速、重复性好等优点,适合献血者抗-HCV的筛查。Objective To investigate the effect of Chemiluminescence immunoassay (CLIA) on detecting the anti-HCV antibody in blood donors and compared with anti-HCV enzyme-linked immunosorbent assay(ELISA) assay and HCV RNA-RT-PCRfluo- rescence quantitative detection assay. Methods We detected anti-I-ICV by CLIA firstly, that the serum specimens of 5 985 do- nors were collected from Jan. 2009 to Aug. 2010. After that the serum specimens treated with reagents 1 and 2 which were two dif- ferent of anti-HCV ELISA reagents as first and repeat assays. All serum positive specimens from anti-HCV ELISA first and repeat assays and CLIA were finally identified by HCV RT-PCR fluorescence quantitative detection. Results Fourteen specimens were positive by CLIA ;while 15 specimens were positive in first assay but 16 specimens were positive in repeat assay by ELISA( 14 specimens were positive in both assays). 13 specimens were positive by HCV-RT-PCR. Conclusion The coincidence rate be- tween HCV CLIA and HCV ELISA was 93.53% in the first assay and 87.50% in the repeat assay. The coincidence rate between HCV CLIA and HCV RNA-RT-PCR fluorescence quantitative detection was 92.86%. The HCV CLIA examination method is sim- ple,quick and repeatability,and can be used to screen blood donors with HCV infection.
