摘 要:从某焦化厂废水好氧生物处理的污泥中分离出一株能以间甲酚为唯一碳源、能源生长代谢的菌株Citro-bacter farmeri。考察了间甲酚在不同初始浓度下Citrobacter farmeri细菌生长动力学,4种等浓度酚类物质的降解先后顺序,分别添加共基质葡萄糖和蛋白胨对低浓度和高浓度间甲酚降解的影响,以及在添加共基质葡萄糖时Citrobacter farmeri降解高浓度间甲酚过程中产生的酶活。结果表明,当间甲酚初始质量浓度低于375 mg/L时,Citrobacter farmeri菌株生长符合一级动力学方程;Citrobacter farmeri降解4种酚类物质的降解顺序依次为苯酚>邻甲酚>间甲酚>萘酚;外加葡萄糖或蛋白胨对质量浓度低于375 mg/L的间甲酚的降解没有影响,但对高于375mg/L的间甲酚降解有明显的促进作用,在最佳葡萄糖添加量为800 mg/L或最佳蛋白胨添加量为640 mg/L时,缩短降解时间分别为30 h和29 h;对酶活的测定发现儿茶酚1,2-双加氧酶和儿茶酚2,3-双加氧酶有明显增大,初步判断Citrobacter farmeri以邻位和间位裂解的途径对间甲酚进行降解。A bacterium named Citrobacter farmeri,which utilizes m-cresol as the sole source of carbon and energy,was isolated from the sludge in the aerobic biological wastewater treatment system of a coking plant.The effects of the Citrobacter farmeri bacteria growth kinetics of m-cresol with various initial concentrations,the degradation sequence of the four kinds of phenols with the same concentration,and the separate addition of glucose or peptone on degradation of m-cresol with low or high concentration were studied.The key enzyme activity related to the ring-cleaving during the high-concentration m-cresol degradation by Citrobacter farmeri was measured to analyze the metabolic pathway when added glucose as co-substrate.The results showed that,the Citrobacter farmeri bacterium growth could be described by first-order dynamic equation when the initial m-cresol concentration was lower than 375 mg/L,the degradation of the four kinds of phenols by Citrobacter farmeri were in the following descending order: phenol,o-cresol,m-cresol,and naphthol.The addition of glucose or peptone had no effect on the degradation of m-cresol with mass concentration lower than 375 mg/L;however,for the m-cresol with concentration higher than 375 mg/L,its degradation could be significantly promoted by glucose or peptone addition;the optimal dosage of the said two reagents were 800 and 640 mg/L respectively,and the shortened degradation time were 30 and 29 h respectively.Through the enzyme activity determination,an obvious enlargement of Catechol 1,2-dioxygenase and Catechol 2,3-dioxygenase was found,which meant that Citrobacter farmeri degrade m-cresol through orth-cleaving and meta-cleaving pathway.
