摘 要:应用PCR-SSCP技术对新疆和田和喀什良种驴群体的DGAT2基因内含子3进行单核苷酸多态性检测和分析,探讨DGAT2基因作为候选基因影响新疆良种驴体尺性状的可能性。结果表明:DGAT2基因存在多态性,其第3内含子有两种等位基因A和B,存在AA和AB两种基因型,没有检测到BB基因型,其中AA基因型为优势基因型。AB型与AA型相比存在碱基发生A→G的突变,使赖氨酸突变为精氨酸。新疆和田良种驴AA与AB基因型个体相比,体高和胸围差异极显著(P<0.01),体长差异显著(P<0.05),AB型体高、胸围和体长高于AA型。新疆喀什良种驴AA与AB基因型个体相比,体高差异显著(P<0.05),AB型的体高、胸围和体长高于AA型。新疆喀什良种驴的体高、体长和胸围高于和田良种驴,差异极显著(P<0.01)。PCR-SSCP method was applied to determine and analyse single nucleotide polymorphism of intron 3 of DGAT2 gene of fine breed the Hotan and Kashgar donkey in Xinjiang,the possibility of DGAT2 gene was studied as candidate gene of body measurement and slaughter quality.The results showed that amplified fragments existed polymorphism,DGAT2 gene intron had two alleles,A and B,two genotyping AA and AB,but BB was not found,AA is the dominant genotype,the Genotype frequency of which is 0.6964(0.3036).A→G mutation existed in AB,and changed the data encoding.Compared AA genotype with the AB of Hotan donkeys,the body height and chest circumference were very significantly different(P0.01),body length significantly different(P0.05).AA genotype compared with the AB of Kashgar donkeys,body height significantly different(P0.05),body height,chest circumference and body length AB-type were higher than AA.Body height,body length and chest circumference of Kashgar donkeys were higher than Hotan donkeys in experimental groups,the difference was significant(P0.01).
