益气补肾中药对自然流产模型小鼠CD28分子表达的影响

摘　　要：目的:观察益气补肾中药对小鼠母胎免疫的调节作用。方法:以CBA雌鼠×DBA/2雄鼠建立小鼠自然流产模型,将20只CBA孕鼠随机分成四组,每组5只。空白对照组(NS组)予生理盐水灌胃,西药组予环孢菌素A(CsA)灌胃,中药组予益气补肾中药灌胃,中西结合组予中药+CsA灌胃。妊娠14天处死孕鼠,取脾脏与母胎界面组织,用流式细胞分析术(FCM)分析各组孕鼠CD28分子的表达情况。结果:CD28分子在脾脏表达趋势从高到低依次为:生理盐水组、西药组、中西结合组、中药组,各治疗组CD28分子表达均低于NS组(P<0.05);CD28分子在母胎界面上表达趋势从高到低依次为:生理盐水组、中药组、西药组、中西结合组,中西结合组CD28分子表达低于NS组、西药组和中药组(P<0.05);中药组、西药组CD28分子表达均低于NS组(P<0.05)。结论:益气补肾中药对自然流产模型小鼠有下调CD28分子表达的作用趋势。Objective：To study the effects of traditional Chinese medicine（TCM） with function of invigorating Qi and toni-fying kidney on the expression of CD28 at spleen and maternal-fetal interface in abortion-prone mice during pregnancy.Methods：CBA female mice were mated to DBA /2 male mice to establish an abortion-prone model.The female micewere divided randomly into 4 groups（5 in each）：control group（NS group）：drenched normal saline everyday since the firstday of gestation;western medicine（WM） group（cyclosporine group）：drenched cyclosporine solution once on the 4th day ofgestation;TCM group：drenched the TCM decoction with the function of invigorating Qi and tonifying kidney everyday since the first day of gestation;and TCM-WM（TCM decoction＋cyclosporine）group：drenched the TCM decoction every-day since the first day and cyclosporine solution on the 4th day of gestation.On the 14th day of gestation,the mice werekilled in batches and their spleen and maternal-fetal interface tissues were made into single cell suspension to detect theexpression of CD28 by flow cytometry（FCM）.Results：The expression of CD28 in spleen tissues in NS group,cy-closporine group,TCM-WMgroup,and TCMgroup was from high to lowin order and it was significantly lower in the lat-ter 3 groups as compared with the NS group（P0.05）.The expression of CD28 at maternal-fetal interface in the NS,TCM,cyclosporine,and TCM-WM group was from high to low in order,with a significant difference between the TCM-WM group and the NS,TCM and cyclosporine groups（P0.05） and between the TCM and WM groups and the NS group（P0.05）.Conclusion：TCM with function of invigorating Qi and tonifying kidney may reduce the expression of CD28 in abortion-prone mice.
