摘  要:以石斛兰单株为外植体,研究了谷氨酰胺、氯化镧对石斛兰离体增殖及叶片POD、CAT同工酶表达的影响。结果表明,随着处理浓度、处理时间、外源添加物的不同,石斛兰增殖指标、叶片POD同工酶表达类型及活性差异明显,而CAT同工酶仅表现活性的差异。其中,处理4与处理5表现出截然相反的结果。随着处理时间的延长,特别是60d时,处理4的组培苗全部死亡,而处理5依然保持了100%最高的外植体增殖率和4.36最大的平均增殖苗数,而POD不仅表现在活性很高,且在B区比处理4多3条酶带。说明,0.05mg.L-1的氯化镧处理40d,不仅有利于石斛兰组培苗的增殖,也能够保证组培苗的质量。By using single plant as explant,effects of glutamine,lanthanum chloride on proliferation of Dendrobium in vitro and POD,CAT isozyme expression of leaves were studied.The results showed that,along with different treatment concentration,processing time and foreign additives,proliferation indexes of Dendrobium and isozyme expression activity,type of POD behaved obvious difference,while CAT isozyme only showed activity difference.Among those treatments,treatment 4 got the opposite results to treatment 5.With the extension of time,especially in the 60days,cultured seedlings of treatment 4 were all dead,on the contrary,treatment 5 remained 100%,the highest rate of explant proliferation and 4.36,the largest proliferation number per explant,its POD activity was much higher than treatment 4 and had 3 more bands in area B.Those indicated 0.5 mg·L-1 treatment for 40 days was not only conducive to the proliferation of Dendrobium,but also was able to guarantee the quality of seedlings.
