摘 要:对鸡传染性支气管炎病毒(IBV)陕西分离株SX-H09株膜蛋白(M)基因进行克隆和序列分析。根据GenBank发表的IBV基因序列设计1对特异性引物,RT-PCR扩增IBV M基因片段,并进行克隆、序列测定和分析。结果表明,SX-H09株M基因全长为678 bp,编码225个氨基酸。同源性比较可知,SX-H09株M基因与参考株M基因的核苷酸序列同源性为82.6%~99.6%,推导的氨基酸序列同源性为81.0%~99.6%。系统发育进化树表明,SX-H09株与参考株DY07和IBVSX7在同一个分支上,亲缘关系较近。In order to provide scientific and effective theoretical basis for the study of the epidemiology of avian infectious bronchitis virus(IBV) and the design of genetically engineering vaccine of IBV,the membrane(M) protein gene of IBV SX-H09 strain isolated from Shaanxi was amplified,and the sequence was analyzed as well as the molecular biology and genetic variation of M gene were investigated.A pair of specific primers were designed according to the gene sequence of IBV published in GenBank,which was used to amplify the fragment of M gene by RT-PCR method.The M gene of IBV SX-H09 contains 678 bp,which encodes a polypeptide of 225 amino acids.Compared with the reference strain,the nucleotide and deduced amino acid sequences were 82.6%-99.6% and 81.0%-99.6% respectively.The phylogenetic tree showed that there was a close genetic relationship between the DY07 and IBVSX7.The result indicated that there was genetic variation in SX-H09 strain.
