摘  要:目的观察低压缺氧对兔动脉粥样硬化斑块低氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)及基质金属蛋白酶-9(MMP-9)表达的影响。方法将日本大耳兔随机分为常氧对照组和低压缺氧组,高胆固醇饮食12周建立兔动脉粥样硬化(atherosclerosis,AS)病变模型后,取主动脉内膜组织分别采用免疫组织化学方法和real-time PCR检测斑块组织中HIF-1α、VEGF、MMP-9蛋白和mRNA表达。结果低压缺氧组高斑块中HIF-1α、VEGF、MMP-9蛋白和mRNA的表达明显升高,与常氧对照组比较,差异有统计学意义(P<0.05)。结论低压缺氧能明显升高斑块中HIF-1α、VEGF、MMP-9蛋白和mRNA表达,可降低兔动脉粥样硬化斑块的稳定性。Objective To observe effects of periodicity hypoxia on contents of hypoxia inducible factor-1(HIF-1α),vascular endothelial growth factor(VEGF)and matrix metalloproteinase-9(MMP-9)in atherosclerotic plaques.Methods Rabbit atherosclerotic models were set up by hypercholesterolemia diet.Contents of HIF-1α,VEGF and MMP-9 protein and mRNA in plaque were detected by immunohistochemistry and Real-time PCR methods,respectively.Results In periodicity hypoxia groups,HIF-1α,VEGF and MMP-9 protein and mRNA in atherosclerotic plaques were increased markedly.Conclusion Periodicity hypoxia could increase contents of HIF-1α,VEGF and MMP-9 protein and mRNA in rabbit atherosclerotic plaques.This maybe decreased stability of atherosclerotic plaques.
